The viral portion of the insert is 8.8 kb. The insert also includes cellular sequences derived from Mo (T cell leukemia) cells. The insert contains the following restriction sites (approximate kb from 5' end): EcoRI--0.1, 0.8, 4.7, 9.0; KpnI--1.7, 3.4; NheI--1.0, 6.65; SacI--0.5, 4.0, 8.7; XbaI--3.6; BamHI--0.3, 5.0, 8.4. The full-length plasmid is unstable in bacterial hosts, and is distributed as purified plasmid DNA. Constructed by inserting the HindIII fragment from lambdaH6 into the HindIII site of pBR322 that had been modified by an EcoRV-MscI deletion of approximately 1.25 kb. Restriction digests of the clone give the following sizes (kb): EcoRI--5.2, 4.0 (doublet), 0.92, 0.36; HindIII--10.5, 3.2; XbaI--13.5. |
