Restriction digests of the clone give the following sizes (kb): EcoRI--3.0, 0.75; BamHI--3.8; HindIII--3.8; PstI--3.8; XbaI--3.8. The insert includes the complete coding sequence, extending from 3 bp upstream of the initiation methionine to 276 bp downstream from the stop codon. The nucleotide sequences encoding the NH2-terminal leader peptide and first 26 amino acids of GYPA and GYPB are nearly identical. There is also a very homologous region encoding the presumed membrane-spanning portion of the proteins. This probe detects the same polymorphism as HGpA-3. |
